Il romanzo è ambientato durante l'assedio di Richmond della Guerra di secessione americana: cinque prigionieri (un ragazzo, un giornalista, un marinaio, un ingegnere, un ex schiavo e perfino un cane) decidono di fuggire dalla città in una maniera piuttosto inusuale rubando un pallone a gas.
Dopo numerosi giorni di volo in balia di una tempesta, il gruppo naufraga su un'isola vulcanica che decidono di chiamare "isola Lincoln", in onore del presidente americano.
Il mistero dell'isola da cui il titolo del nostro è dovuto ad alcuni avvenimenti inesplicabili:come uno dei fuggitivi sia riuscito a sopravvivere alla caduta dal pallone .. il salvataggio del cane da una bestia marina...il ritrovamento di una cassa piena di beni di necessità..il rinvenimento di un messaggio in mare richiedente soccorso..
L'isola misteriosa costituisce il terzo e ultimo capitolo di una trilogia iniziata con "I figli del capitano Grant "e "Ventimila leghe sotto i mari ".
Vi si ritrovano pertanto alcuni personaggi dei due capitoli precedenti e allusioni alle vicende in essi narrate.

L’autore di queste indicazioni è Arne Saknussem, alchimista islandese, che nel XVI secolo avrebbe davvero compiuto l’impresa. Londra, 1872 . Il ricco inglese Phileas Fogg è solito frequentare il Reform Club.
Un giorno si accende una discussione in merito ad un articolo riguardante la costruzione di una nuova linea ferroviaria in India: il giornale riporta che sarebbe possibile a quella velocità effettuare il giro del mondo in 80 giorni.
Tutti ne mettono in dubbio la possibilità, ma il protagonista decide di scommettere 20.000
sterline: entro quel lasso di tempo Fogg ed il suo servo Passepartout dovranno essere di
ritorno al club dopo aver girato il mondo.
I due si mettono in viaggio verso Bombay e giunti in India, scoprono che la linea ferroviaria in realtà non è ancora stata ultimata e ciò causa loro una grossa perdita di tempo. Con un ispettore di Scotland Yard (Fix) alle calcagna ed una bella ragazza indiana da salvare, Fogg fa prima tappa ad Hong Kong e poi in Giappone, alla volta della nave per San Francisco. Arrivati a New York, i protagonisti scoprono che l’imbarcazione per Liverpool è già partita.Sembra tutto perduto, ma Passepartout avrà più di un'intuizione geniale per tornare al Reform intempo e vincere la scommessa !
